目的

探讨大鼠急性脊髓损伤(SCI)后损伤段脊髓组织中是否存在赖氨酰氧化酶(LOX)的表达及其意义。

方法

将70只健康SD雌性大鼠分为A、B、C三组,A组为单纯损伤组,B组为氯化钴组,C组为正常对照组,采用改良式Allen氏打击法制成大鼠急性脊髓损伤模型,A、B组分别在损伤后1 d、3 d、7 d、14 d四个时间段分别处死大鼠,取损伤区脊髓组织制成石蜡切片,采用免疫组织化学法检测LOX在脊髓损伤组织中的表达,并分析A组、B组与C相比之差异性,同时分析A组、B组不同时间窗的相关性。统计学分析方法,分析A组、B组与C相比之差异性,同时分析A组、B组不同时间窗的相关性。实验数据采用SPSS17.0统计软件包进行处理,所有数据以均数±标准差(±s)表示,组间比较采用单因素方差分析(ANOVA)及t检验,P<0.05为差异具有统计学意义。

结果

与正常对照组(C组)相比,单纯脊髓损伤组(A组)1d LOX阳性表达明显增加,3 d表达比1 d略微减少,7d时达到最高峰,14 d时明显减少。正常对照组(A组)与单纯损伤组(B组)中LOX的表达分别为4.96%和41.43%(t=4.602,P=0.006<0.01)差异具有统计学意义;氯化钴组(B组)与单纯损伤组(A组)LOX阳性表达曲线基本相同,但阳性表达率却比A组升高,为71.30%(t=2.3055,P=0.028<0.05)差异具有统计学意义。1 d LOX阳性表达B组比A组升高(t=5.208,P=0.002<0.05),3 d LOX阳性表达B组比A组升高(t=3.142,P=0.021<0.05),7 d LOX阳性表达B组比A组升高(t=3.007,P=0.028<0.05),14 d LOX阳性表达B组比A组升高(t=3.246,P=0.014<0.05),相同时间段间两两比较都具有统计学意义。

结论

SCI后受损段脊髓细胞出现LOX的表达并逐渐开始升高,与继发性SCI缺血缺氧密切相关,其表达规律与损伤时间具有相关性。

Objective

Toexplore the expression of lysyl oxidase(LOX)and its significance in the damaged spinal cord tissue sections of rats after acute spinal cord injury.

Methods

70 healthy SD female rats were divided into three groups A,B,C,A for pure damage group,B group of cobalt chloride group,group Cas normal control group,A for pure damage group,B group of cobalt chloride group,group C as normal control group,the rats in the A,B group were sacrificed respectively at four times of 1 d,3 d,7 d,14 d after injure,the injured area of spinal cord tissue was made of paraffin,the expression of LOX was detected by immunohistochemistry in tissue of spinal cord injury,and analyzed the differences between A group and B group analysised the correlation between A and B group of different time periods at the same time.Analyze the differece of groups A,B and C by Statistical analysis methods,correnlation analysis of group A and group B in different time windows.Experimental data using SPSS17.0 statistical software package for processing,all data were expressed as(±s)indicate,group were compared using ANOVA and t test,P<0.05 was considered statistically significant.

Result

Compared with normal control group,expression of LOX increased in pure damage group at the 1 d,decreased slightly at 3 d,reached the peak at 7 d,then significantly reduced at 14 d.The normal control group(group A)and the simple injure group(group B)expression of LOX were 4.96% and 41.43% (t=4.602,P=0.006<0.01)difference was statistically significant;Cobalt chloride group(group B)with simple injure group(A)LOX expression is basically the same curve,however,the positive expression rate higher than the in group A,was 71.30%(t=2.3055,P=0.028<0.05)difference was statistically significant.1d positive expression of LOX in B group than in A group increased(t=5.208,P =0.002<0.05),3 d positive expression of LOX in B group than in A group increased(t=3.142,P=0.021<0.05),7 d positive expression of LOX in B group than in A group increased(t=3.007,P=0.028<0.05),14d positive expression of LOX in B group than in A group increased(t=3.246,P=0.014<0.05),at the same time each other compared with statistical significance.

Conclusion

Damaged segment of spinal cord cells appear the expression of LOX and gradually began to rise after SCI,which is closely associated with secondary SCI ischemia hypoxia,and there is a correlation between its expression pattern and injury time.
