目的

观察皮肤涂抹火箭煤油对二硝基氟苯(DNFB)所致小鼠迟发型过敏反应(DTH)的影响。

方法

纳入雌性120只ICR小鼠,按照小鼠体质量随机分组,每组6只。不同剂量火箭煤油1次染毒(1 d,1次/d)分为阴性对照组、阳性对照组、100 μL组、200 μL组、400 μL组;根据100 μL同一剂量火箭煤油不同次数染毒(不同天数,1次/d):分为阴性对照组、阳性对照组、1 d、2 d、3 d、4 d、5 d组;不同剂量火箭煤油多次染毒(5 d,1次/d):分为阴性对照组、阳性对照组、100 μL组、200 μL组、400 μL组。小鼠背部剃毛,阴性对照组和阳性对照组涂抹丙酮,各剂量试验组涂抹相应剂量的火箭煤油。除阴性对照组外,每鼠背部用1%DNFB溶液均匀涂抹进行致敏,次日强化,致敏后第5天,将1%DNFB溶液均匀涂抹于所有组小鼠右耳进行攻击,攻击后24 h,测定右耳的肿胀度,脾指数和胸腺指数。

结果

不同剂量火箭煤油单次染毒时(1 d),与阳性对照组对比,200 μL ×1 d组小鼠耳肿胀度和肿胀率显著降低,胸腺指数显著升高(bt胸腺指数=2.27,P<0.05);400 μL×1 d组的小鼠耳肿胀度和肿胀率显著降低,胸腺指数显著升高(bt肿胀度=28.07、bt肿胀率=26.96、bt胸腺指数=5.71,P<0.05),随染毒剂量的增加,变化趋势更为明显;与阳性对照组比较,100 μL×5 d组的小鼠耳肿胀度、肿胀率和脾指数显著降低,胸腺指数显著升高(bt肿胀度=8.23、bt肿胀率=4.51、bt胸腺指数=5.29,P<0.05),且随染毒次数的增加,变化趋势更为明显;不同剂量航天煤油多次染毒时(5 d),与阳性对照组比较,100 μL组、200 μL组和400 μL组小鼠耳肿胀度、肿胀率、脾指数显著降低,胸腺指数显著升高(100 μl×5组:bt肿胀度=868.65、bt肿胀率=10.35、at脾指数=3.55、bt胸腺指数=2.69,P<0.05;200 μl×5组:bt肿胀度=55.53、bt肿胀率=34.67、at脾指数=4.28、bt胸腺指数=2.81,P<0.05;400 μl×5组:bt肿胀度=81.69、bt肿胀率=49.90、at脾指数=4.65、bt胸腺指数=5.37,P<0.05),而随染毒剂量的增加,呈现显著的变化趋势。火箭煤油染毒剂量增大和染毒次数增加抑制DNFB所导致的致敏作用程度增加。

结论

火箭煤油可抑制DNFB诱导小鼠DTH,具有细胞免疫毒性。

Objective

To determine the effect of rocket kerosene on delayed type hypersensitivity (DTH) by dinitrofluorobenzene (DNFB) in mouse.

Methods

The first series of experiments included that different dosage rocket kerosene was applied once; ICR mice were randomly divided into the negative control group, positive control group and experimental group (100 μL×1, 200 μL×1 and 400 μL×1 group). The second series of experiments included that same dosage rocket kerosene was applied different times; ICR mice were randomly divided into the negative control group, positive control group and experimental group (100 μL×1, 100 μL×2, 100 μL×3, 100 μL×4 and 100 μL×5 group) . The third series of experiments included that different dosage rocket kerosene was applied more than once. ICR mice were randomly divided into the negative control group, positive control group and experimental group (100 μL×5, 200 μL×5 and 400 μL×5 group). Different amounts of the undiluted fuel were applied directly to the dorsal skin of the mice. Every experimental control groups were treated with acetone. After the last exposure, all the mice were smeared evenly with 1% DNFB solution on the dorsal for allergy except the negative control group and in the same way on the second day. Five days after allergy, 1% DNFB solution was smeared to right ear of all mice to stimulate the allergic reaction. The auricle swelling, spleen index and thymus index in right mouse were determined 24 hours after attack.

Results

When different dosage of rocket kerosene were applied once (1 d), compared with positive control group, the auricle swelling decreased and the thymus index increased significantly in 200 μL×1 group (btthymus index=2.27, P<0.05); the auricle swelling decreased and the thymus index increased significantly in 400 μL×1 group (btauricle swelling index=28.07, btauricle swelling rate=29.96, btthymus index=5.71, P<0.05). When different dosage of rocket kerosene were applied different times (5 d), compared with positive control group, the auricle swelling, spleen index decreased and thymus index increased significantly in experimental group (100 μL×5 group: btauricle swelling index=868.65, btauricle swelling rate=10.35, btspleen index=3.55, btthymus index=2.69, P<0.05; 200 μL×5 group: btauricle swelling index=55.53, btauricle swelling rate=34.67, btspleen index=4.28, btthymus index=2.81, P<0.05; 400 μL×5 group: btauricle swelling index=81.69, btauricle swelling rate=49.90, btspleen index=4.65, btthymus index=5.37, P<0.05). Different dosages of rocket kerosene benefited to reduce auricle swelling, spleen index and elevate thymus index in mouse with allergic reaction induced by DNFB at different degree.

Conclusion

Rocket kerosene can inhibit the delayed type hypersensitivity in mouse induced by DNFB and it has cell mediated immunotoxicity.
