目的

探讨心肺复苏对室颤大鼠脑神经元线粒体功能的保护作用。

方法

通过电刺激致颤法建立大鼠心室颤动/心肺复苏模型,随机分为对照组,缺血组,CPR组。用Clark氧电极法检测线粒体呼吸功能;电镜观察线粒体超微结构;液氮冻存海马神经元线粒体通过效液相色谱检测磷酸肌酸(PCr)、三磷酸腺苷(ATP)和乳酸(Lac)含量。

结果

CPR组大鼠脑线粒体态III呼吸和RCR值显著降低。电镜下三组大鼠神经元线粒体超微结构无明显差异。缺血组和CPR组PCr和ATP含量均明显小于对照组(P<0.01)。

结论

心跳骤停引起脑线粒体呼吸功能严重损伤,心肺复苏可以改善神经元的线粒体功能,减轻脑线粒体损伤。

Objective

To investigate the protective effect of cardiopulmonary resuscitation on brain mitochondrial function in a rat model of cardiac arrest.

Methods

Ventricular fibrillation(VF) was electrically induced via alternating current delivered to the right ventricular endocardium. The rats were randomized into three groups, ie, control group, ischemia group and CPR group. Clark oxygen electrode was used to measure mitochondrial respiratory function. Mitochondrial ultrastructure was observed under transmission electron microscope. Hippocampal mitochondria was removed rapidly and quick-frozen in liquid nitrogen for further HPLC analysis.

Results

The levels of State Ⅲ O2 consumption and RCR in CPR group were significantly less than that in control group. There was no morphological difference in between two groups. The content of PCr and ATP in both ischemia group and CPR group were less than that in control group.

Conclusions

Cardiac arrest causes severe injuries in brain mitochondrial respiratory function. CPR could improve mitochondrial function and alleviate ischemic neurons.
