Effects of melatonin on oxidative injury in lungs of mice induced by inhalation exposure to rocket

Abstract:

Objective

To observe the protective effects of melatonin on oxidative injury in lungs of mice by inhalation exposure to rocket kerosene.

Methods

18 ICR male mice were divided into normal control group,rocket kerosene(RK)group and melatonin(MT)group.The normal control group were received no treatment.The RK group mice were administered i.g.0.5% sodium carboxymethyl cellulose 30 min before inhalation exposure.The melatonin group were administered i.g.melatonin(2 mg/kg)30 min before inhalation exposure.RK group mice were inhaled with kerosene in 18 000 mg/m3 and control group were inhaled with fresh air for 4 hours.Oxidative Stress was assessed by assay of MDA,SOD,GSH-PX and TAOC.

Results

Compared with the NCG MDA:(0.98±0.15)nmol/mg Pr;TAOC:(1.33±0.11)U/mg Pr;SOD:(5.21±0.38)U/mg Pr;GSH-Px:(73.26±12.69)U/mg Pr,RK inhaltion produced a signi?cant increase in the lung MDA levels(1.18±0.04)nmol/mg Pr and produced a signi?cant decrease in the TAOC(1.19±0.10)U/mg Pr,SOD(4.40±0.29)U/mg Pr and GSH-Px(56.83±9.70)U/mg Pr activity after 2 hour post-exposure(P<0.05).Compared with RK group,MT group produced a significant decrease in lung MDA levels(1.07±0.11)nmol/mg Pr,P<0.05 and produced a significant increase in the TAOC(1.29±0.06)U/mg Pr,SOD(4.93±0.44)U/mg Pr and GSH-Px(67.17±3.67)U/mg Pr activity after 2 hour post-exposure(P<0.05).

Conclusion

Melatonin can reduce oxidative stress,and have some protective effects on lung oxidative injury induced by inhalation exposure to rocket kerosene.
